In vitro Propagation of Hyacinthus orientalis L.




Abstract
The study was carried out in order to propagate efficiently bulb of hyacinth to use Sower stalk tissue as culture materials with results clarified through the experiments on comparison of totipotency between flower stalk with inflorescence and one without those, determination of optimal concentration for bulblet formation, difference between organogenesis capability between light or dark condition and test of efficiency of bulblet differentiation from bulbscale and flower stalk tissue of Hyacinthus orientalis. The results obtained were summarized as follows.
Differentiation of bulblet from flower stalk tissue with inflorescence was better, compared with that without inflorescence in all the varieties tested. Percentage of bulblet formation and proliferation of bulblet of $quot;Carnegie$quot; were best in 1.0§·/§¤ IBA and that of $quot;Anna marie$quot;, in 1.0§·/§¤, IAA but growth of bulblet of $quot;Carnegie$quot; was good in 0.1ppm IAA, that of $quot;Anna marie$quot;, in 1.0§·/§¤ IAA added medium.
Bulblet formation percentage of $quot;Lady derby$quot; and $quot;Ostara$quot; was higher in the media supplemented with 1.0§·/§¤ IBA, growth of bulblet in those varieties was better in 1.0§·/§¤ IBA added medium of dark culture plot.
Percentage of callus and bulblet formation, and number of bulblets/explant were higher in Sower stalk tissue with inflorescence than in bulb scale tissue of mother bulb, but those were bad in flower stalk without inflorescence. Bulblet formation percentage and number of bulblets/explant from flower stalk tissue was good in 1.0§·/§¤ IBA, while those of bulb scale tissue, in 1.0§·/§¤ IAA added medium.
